Studies on genotype specific modified methods for cryopreservation of cock semen

G.-P. Schramm

In comparison with artificial insemination with fresh semen or short time stored semen, the results of artificial insemination after cryopreservation are not satisfactorily in poultry. Nevertheless, interest of commercial breeders in cryopreservation is increasing. An increase in utilization of these methods for practical poultry reproduction is expected through population specific adapted methods of semen cryopreservation. That can also be a contribution to effective preservation genetic poultry resources. Three different freezing methods using “Blumberg-Cock-Semen-Extender” were analysed. Additional variations were received by different concentration of the cryoprotector dimethylformamide and varying thawing temperatures. Cocks and hens of four breeds were used (White Leghorn, White Meat Type, Fayoumi and Dwarf Rhode Island Red). The results showed clear method dependent effects on the in vitro quality and on fertilization capacity of thawed semen. In addition, the analysis expressed a different genotype specific suitability of the tested methods for cryopreservation of cock semen. By using of such genotype adapted cryopreservation methods, it seems also to be possible to improve the fertilization conservation of frozen semen from cocks having genetically reduced native sperm quality.
